Chemotheraputic Agents Flashcards
List the sulfonamides
Sulfamethoxazole(p450 inhibitor) sulfadiazine and sulfasalazine.
Sulfonamides mechanism of action
Inhibits folate synthesis by inhibiting dihydropteroate
Sulfonamides clinical uses
Nocardiosis, co-trimoxazole for UTI
Sulfonamides AEs
Hyper sensitivity (SJS), photosensitivity, haemolytic anemia in G6PD deficiency, kernicterus in infants and crystalluria
What can Sulfonamides do to warfarin
Displace it from albumin
Can sulfonamides be given to pregnant ladies ?
No they are teratogenic
Trimethoprim mechanism of action
Inhibits dihydrofolate reductase (DHFR)
Trimethoprim clinical use
Used in combination of sulfonamides
Co-trimoxazole—> used for UTI and bacterial prostitis, P.carinii
Trimethoprim AEs
Megaloblastic anemia, leukopenia and granulocytopnia, hyperkalemia
What is co-trimoxazole
Sulfamethoxazole + trimethoprim —> Co-trimoxazole (active against MRSA)
List the fluoroquinolones
Ciprofloxacin (p450 inhib), levofloxacin, moxifloxacin and gemifloxacin
Suffix - floxacin
Fluoroquinolones mechanism of action
Inhibits prokaryotic enzymes topoisomerase (gyrase) 2,3,4 hence inhibiting bacterial DNA synthesis
Ciprofloaxcin which topoisomerase does it inhibit
Topo 2
Levifloxacin and Moxifloxacin which topoisomerase does it inhibit
Topo 4
Ciprofloaxcin topo 2 clinical use
Gram negative P. Arguinosa
