Q

What is Megaloblastic anemia?

A
  • Result of folate deficiency in which blast cells are unable to replicate DNA and divide
  • They just keep on getting bigger leading to weak membrane and poor O capacity

7
Q

What does 5-methyl-THF do?

A
  • Converts homocysteine to methionine
  • Homocysteine is toxic and methionine is used in AA metabolism
  • This is the only reaction 5-methyl-THF is used in and the reaction converts it back to THF
  • B12 required for reaction

16
Q

What drug inhibits purine synthesis?

A

6 mercaptopurine

17
Q

What cancer is 6 mercaptopurine used for?

18
Q

Why do tumor drugs cause increased risk of infection?

A

WBCs formation is inhibited in marrow as well making susceptible to infection

19
Q

What is necessary for pyrimidine synthesis?

A

Co2, glutamine, and aspartate are necessary for synthesis

20
Q

What drug is useful in treating reumatoid arthritis?

A

Leflunomide inhibitis pyrimidine synthesis stopping WBC inflamatory reaction

21
Q

What does Thymidylate synthase do?

A
  • converts dUMP to dTMP
22
Q

Why is Thymidylate synthase good target for cancer drug?

A

Thymine is only in DNA and not RNA

23
Q

What does 5 fluorouracil do?

A

Blocks action of thymidylate synthase

24
Q

What is Dihydrofolate?

A

Product of pyrimidine synthesis that is toxic and needs to be converted by to tetrahydrofolate

25
What does Methotrexate do?
Stops Dihydrofolate reductase which converts DFH to THF poisoning cell as DHF is toxic
26
What does Ribonucleotide reductase do?
- Responsible for formation of deoxyribose | - Good anti tumor target as deoxy is present in DNA but not RNA
