Pyruvate Flashcards
Once pyruvate is made in the cytosol where does it go
the mitochondria
In the mitochondria, what is the first enzyme pyruvate encounters
pyruvate DH turns it into acetyl-coA makes NADH and CO2
Acetyl-coA can be used in
TCA cycle to make more ATP turned into fatty acids
Pyruvate DH is inhibited by
acetyl-coA
What 5 things does pyruvate DH need to work
thiamine pyrophosphate (thiamine) lipolic aicd conenzyme A (pantothenate) FADH2 (riboflavin) NADH (niacin and some from tryptophan)
An alcoholic comes in and you give them IV glucose and they die, why?
lactic acidosis build up of pyruvate no thiamine for pyruvate to move forward goes to lactate instead
What 4 pathways can pyruvate go into
- Made into alanine via alanine aminotransferase (B6), carries amino group to liver from muscle
- Made into oxaloacetate via pyruvate carboxylase(biotin), replenish TCA or used in gluconeogenesis
- Made into acetyl-CoA via PDH (B1, B2, B3, B5, lipoic acid) go to TCA
- Made into lactate via LDH (B3) for anaerobic glycolysis
Lactate production is major pathway in
- RBCs
- WBCs
- kidney medulla
- lens
- testes
- cornea
What does arsenic inhibit that is important to PDH
Lipoic acid needed as cofactor
- pale skin, skin cancer
- vomiting/diarrhea
- QT prolongation
- garlic breath
