Nonhematopoietic Stem Cells Flashcards
Donor lymphocyte infusion
Purpose
Help to achieve long lasting remission after disease relapse after HSCT
Donor lymphocyte infusion
Process
Lymphocytes collected by leukapheresis and aliquoted into doses
Donor lymphocyte infusion
Dosage
Initial dose: typically 1x10^6 CD3+/kg recipient
Doses increase in CD3 count by half log or 1 log increments as much as 3 months apart
Donor lymphocyte infusion
Most efficient for which disease
CML
Donor lymphocyte infusion
Least effective with which disease
ALL
also only 15-40% of AML see response
Possibly due to more rapid proliferation of tumor cells
Donor lymphocyte infusion
Complications
GVHD- possible increased opportunistic infections
Marrow aplasia
Cytotoxic T cell infusion
Virus specific CTLs (CD8+) have successfully treated CMV and EBV (demonstrated to persist as long as 7 yrs in recipient)
NK cell antigen markets
CD3-, CD56+
Sipuleucel-T
Prostate cancer treatment from enriched autologous dendritic cells that have been activated ex vivo with recombinant protein
MSC identification criteria
Adherence to tissue culture plastic
Positive: CD73, CD90, CD105
Negative:CD45, CD34, CD14, CD11b, CD79a, CD19, HLA-DR
Demonstrate in vitro differentiation to osteoblasts, adipocytes, and chondrocytes
MSC HLA matching
Not necessary- do not induce GVHD or get rejected by recipient T cells
