What is multiple myeloma
Neoplasia of plasma cells in BM

MM is more prevalent in whom?

How do plasma cells –> myeloma?
Lots of DNA rearrangement during antibody production.
This can lead to translocations and Chromosomal deletions
If these affect TS genes/oncogenes –> myeloma
Why does myeloma –> bone disease?
MM cells stimulate osteoclasts and inhibit osteoblasts –> osteolytic lesions
What effect does myeloma have on the kidenys
PCT necrosis
Renal tubular acidosis
Cast nephropathy
Features of multiple myeloma -name 5
Paraproteinaemia Calcium - high Renal failure Anaemia Bone - pain, osteolytic lesions

Why are multiple Tx needed for MM?
Clone cells undergo proliferation but these compete with each other. Different sub clones respond to diff treatments

steroids
proteasome inhibitors
How does autoSCT work
Take blood from patient
Treat with mephalan - to kill MM cells
Reinfuse with treated blood
