Antipsychotics (Typical) Reserpine MPTP (methylphenyltetrahydropyridine)
Class: Antiparkinsonism
MOA: Dopamine precursor. Inhibits peripheral metablosim via dopa decarboxylase
Uses: DOC of Parkinson’s disease
SE: On-off phenomena, Wearing off phenomena
CI: Px w/ Psychosis
Levodopa-Carbidopa
Parkinsonian Phenomena
- alternating periods of improved mobility and akinesia, occuring over a few hours to days during treatment
On-off phenomena
Parkinsonian Phenomena
- Deterioration of drug effect in between meication doses
- Due to progressive destruction of nigriostriatal neurons that occurs w/ disease progression
Wearing off phenomena
Class: Antiparkinsonism drug
MOA: Partial agonist at D2 receptors
SE: Erythromelalgia, pulmonary fibrosis
Bromocriptine
Class: Antiparkinsonism drug
MOA: Partial agonist at dopamine D3 receptors in brain
SE: Behavioral changes, Compulsive gambling, hypersexuality, over eating
Pramipexole
Class: Antiparkinsonism drug
Use: Off-periods of parkinson’s disease, alcoholism
Apomorphine
Notes: Premedicate with Trimethobenzamide to prevent nausea
Class: Antiparkinsonism drug
MOA: Selective inhibitors of monoamine oxidase type B

Class: Antiparkinsonism drug
MOA: Block L-dopa metabolism by inhibiting COMT
Use: Parkinson’s disease (wearing-off phenomena)
SE: Sleep disturbance, orange urine, hepatotoxicity (Tolcapone only), Neuroleptic malignant syndrome, rhabdomyolysis
