DNA Extraction Lab Flashcards
3 basic steps in DNA Extraction
- cell must be lysed( broken open) to release the nucleus
- Nucleus is open to release DNA
- Once the DNA is released it must be precipitated out of the solution
reagents
a substance or mixture for use to produce a chemical reaction
reagents required
- salt-
- detergent-breaks down lipid membrane-
- alcohol–ethanol precipitates the dna
both cell and nuclear membrane are made mostly of what macromolecule
lipids (phospholipid)
In order for the cell to be lysed what must first occur
The lipid membrane must first be broken down
What reagents are used to break down lipid membranes of cells (. How?
- Manual grinding
- detergent solution
Soap molecules mix with fats or lipids, causing structures made if lipids to break apart
alcohol–ethanol
precipitates the dna
Salt
It provides the Dna with a favorable environment by contributing POSITIVELY CHARGE atoms that neutralizes the normal negative charge of DNA, allowing it to clump together
the charge of DNA
NEGATIVE ( also pole/soluble)
