Heat fix
allows the specimen or cells to stick to the slide, that way when staining afterwards the cells won’t wash them away. over heat fixing can kill the cells
Simple staining
staining yeast cells with methylene blue
Differential stains
use multiple stain that distinguish different microbial groups or structures and typically require a more complex procedure than simple stain
examples of differential stains are what?
gram staining, acid fast staining, and endospore staining.
Acid-fast staining
used to identify species that contain mycolic acids and other complex lipids in their cell walls.
Endospore stainning
distinguishes endospores in sample from the vegtative cells
postive stains
where the dyes used color the specimen
negative stains
instead colors the background by adhering to the slide instead of the specimen, creating a silhouette against a darkbackground
