The Effect Of PH on Enzyme Activity Flashcards
Procedure
Put a drop of iodine solution into every well in the spotting tile
Place a Bunsen burner on a heat proof mat and a tripod and gauze over the bunsen burner
Place a beaker of water over the flame until it is around 35 degrees using a thermometer .
Use a syringe to add 1 cm3 of amylase solution and 1cm3 of buffer solution with a PH of 5 to a boiling tube . Place the tube into the hot water
Use a different syringe to add 5 cm3 of starch solution to the boiling tube
Mix the contents of the tube and start a stop clock
Take a fresh sample of the water every 30 seconds and put a drop of it onto the well of the spotting tile
When the Iodine remains a browny orange the starch is not present
Repeat the experiment with buffer solutions of a different PH value
How do you calculate rate of reaction
1000/ time
