Leuprolide mechanism
GnRH agonist
Ethinyl estradiol, DES (causes clear cell carcinoma), mestranol mechanism
bind and stimulate estrogen receptors
Contradications to estrogen therapy
ER+ breast cancer, history of DVTs
Clomiphene mechanism
Hypothalamic estrogen receptor antagonist. Increases release of FSH and LH to stimulate ovulation (treats infertility)
Adverse effects of clomiphene
hot flashes, ovarian enlargement, multiple pregnancies, visual disturbances
Treatment of ER+/PR+ breast cancer but can increase risk of endometrial hyperplasia/carcinoma
Tamoxifen
Treatment for osteoporosis
Raloxifene (ER agonist at bone and antagonist at breast and uterus)
Adverse effect of Raloxifene
increased risk of thromboembolic events
Treatment of ER+ breast cancer in postmenopausal women
Anastrozole, Letrozole, exemestane (aromatase inhibitors)
Use of Terbutaline and Ritodrine
beta-2 agonists that relax uterus and decrease contraction frequency of women in labor
Treatment for hereditary angioedema
Danazol
Adverse effects of danazol
weight gain, edema, hirsutism, decreased HDL, hepatotoxicity
Nonsteroidal competitive inhibitor at androgen receptors used to treat prostate carcinomas
Flutamide
Inhibits steroid synthesis (17,20 desmolase), treats PCOS
Ketoconazole
Treatment for PCOS by inhibiting steroid binding, 17alpha hydroxylase and 17,20 desmolase
Spironolactone
