Oestrogen actions
Prevents ovulation as suppresses the HPO axis and LH so controls menstrual cycle
Stimulates growth of endometrium and breasts
stimulates PR production
Increase HDL and LDL
Sodium and water retention
impair glucose intolerance
Oestrogen uses
control menstrual cycle to prevent pregnancy and ovulation
decrease menopause side effects
Oestrogen side effects
Endometrial, Breast and ovarian hyperplasia
Breast tenderness, nausea, vomiting, water retention, thromboembolism, impaired glucose tolerance
Interacts with Anti-epileptics, st johns wort as all produce CYP450
Progesterone examples
Progesterone depot, implant, low dose progesterone, with oestrogen
Medroxyprogesterone, Dydrogesterone, Nothiesterone, Levonogestrel, Desogestrel
Progesterone actions
Secretory endometrium, increase bone mineral density, mood changes, maintains pregnancy, metabolised by CYP450 enzymes in liver, prevents ovulation by suppressing HPO axis and LH
Progesterone uses
control menstrual cycle to prevent pregnancy and ovulation
decrease menopause side effects
Progesterone side effects
Weight gain, fluid retention, nausea, vomiting, acne, irritability, depression

Clomiphene
Tamoxifen
SERM mechanism of action
Competes with oestrogen for ER binding
Increase anterior pituitary hormones and causes ovulation
Tamoxifen- ER antagonist in breast and causes cell o arrest cell cycle
SERM uses
Clomiphene- treats anovulation
Tamoxifen- treats and prevents breast cancer
