Pharm 38 Cancer Drugs Mechanisms Flashcards
Mechanism: Inhibit thymidylate synthase
- Fluorouracil (5-FU), Capecitabine, Pemetrexed
- (FluCapPem)
Inhibitors of Purine Metabolism
- 6-Mercaptopurine, Azathioprine, Pentostatin
- 6-MP and AZA inhibit IMPDH (Inosine Monophosphate Dehydrogenase) while Pentostatin inhibits adenosine deaminase (ADA)
Inhibits Ribonucleotide Reductase
Hydroxyurea
Purine Analogues
Thioguanine (Guanine), Fludarabine Phosphate (Purine), Cladribine (Adenosine)
Pyrimidine Analogues
- Cytarabine (araC), Azacytidine, Decitabine, Gemcitabine (Bines and Dine)
- All are Cytidine Analogues
Alkylating Agents (Covalently bind DNA)
Cyclophosphamide, Mechlorethamine, Melphalan, Estramustine
Modify DNA Structure by Cross-linking intrastrand guanine bases
Cisplatin, Carboplatin, Oxaliplatin
- (Platinum Compounds)
Binds oxygen and chelates Fe(II); binds DNA and leads to strand breaks via generation of oxidative intermediates
Bleomycin
Inhibit Topoisomerase I, leading to DNA strand breakage
Irinotecan, Topotecan (Tecans Block Type I)
Inhibit Topoisomerase II, leading to DNA strand breakage
- Doxorubicin, Daunorubicin, Epirubicin, Etoposide, Teniposide, Amsacrine
- Rubicins, Posides, and Amsacrine block Topo II
Bind tubulin subunits and prevent microtubule polymerization (Bind to + end)
- Vinblastine, Vincristine, Eribulin (Vins and Bulins bind Tubulin)
Bind polymerized tubulin and inhibit microtubule depolymerization
Paclitaxel, Abraxane, Docetaxel (Taxels and Xane)
