Antiemetic

Mechanism of Action:
5-HT3 (serotonin) R antagonist ⇒ based on the theory that 5-HT3 receptors are present at the vagal nerve terminals and binding of serotonin initiates vomiting reflex, Zofran would prevent this process

Onset of Action = Est. 10-30 minutes

Half-Life = 4 hours

Indications/Field Use:
- Nausea
- Vomiting
ONDANSETRON (Zofran®)
Contraindications:
Hypersensitivity
ONDANSETRON (Zofran®)
Side Effects/Adverse Reactions:
- HA
- dizziness
- tachycardia
- chest pain
- constipation
- diarrhea
- dry mouth
- prolonged QT interval

ADULT DOSE:
(ADULT): 4mg slow IVP (IO) over 2-5 min

PEDI DOSE:
(PEDI): 0.1mg/kg slow IVP (IO) over 2-5 min

Precautions:
Lower risk of sedation and dystonia

than other antiemetics

Drug-Drug Interactions:
May affect clearance and half-life with inducers and inhibitors of Cytochrome P-450 enzymes (CYP3A4, CYP2D6, CYP1A2)
