A proton pump inhibitor (PPI)

What are the common indications for Omeprazole?
A
- remove H. pylori
- gastric or duodenal ulceration
- GORD
- Zollinger-Ellison syndrome
- Oesophagitis
- Dyspepsia

What is the mechanism of action of Omeprazole?
A
Inhibits the proton pump, H+-K+-ATPase pump (final common pathway)
Inhibits irreversibly

What are the common adverse effects of Omeprazole?
A
- GI disturbance - abdominal pain, constipation, diarrhoea
- Headache, dizziness
- Drowsiness, confusion

What are the common drug-drug interactions of Omeprazole?
A
Omeprazole is a CYP inhibitor so reduces the action of clopidogrel (a pro-drug activated by these CYP enzymes)
Can increase the effects of warfarin and phenytoin so need to monitor
Often prescribed alongside NSAIDs and steroids due to their risk of ulceration
