What drug class is Co-careldopa?
A
Levodopa (dopamine precursor) + peripheral DOPA decarboxylase inhibitor

What are common indications for Co-careldopa?
A
- Parkinson’s disease

What is the mechanism of action of Co-careldopa?
A
The L-DOPA is converted to dopamine in the brain (L-DOPA can cross the BBB) but it can be inactivated in the periphery by DOPA decarboxylase, so if you give it with an inhibitor of this, it prevents it’s degradation

What are the adverse reactions of Co-careldopa?
A
Fewer side effects than L-DOPA
- irregular HR
- Nausea and vomiting
- Headache
- SOB
- Chills
- Dyskinesia
Long term, the drug wears off

What are the common DDIs of Co-careldopa?
A
- Vitamin B6 increases peripheral breakdown of L-DOPA
* Antipsychotic drugs - block dopamine receptors and lead to parkinsonism side effects
