Selective COX-2 inhibitor (NSAID)

What are the common indications for Celecoxib?
A
- Osteoarthritis

What is the mechanism of action of Celecoxib?
A
Inhibits COX-2 - inhibits PGI2 and PGE2 production
However unlike COX-1 inhibitors, it doesn’t inhibit thromboxane A2 production
(Etorcoxib is more selective than Celecoxib)

What are the common adverse reactions of Celecoxib?
A
- Renal ADRs (Reduced GFR and reduced renal blood flow) - therapeutic doses, less adverse reactions
- less GI ADRs as more selective (dyspepsia, GI bleeding, nausea, peptic ulceration)
- Risk of MI

What are the common drug-drug interactions of Celecoxib?
A
- Aspirin
- Glucocorticoid steroids
- Anticoagulants (give with PPI)
- ACEi
- ARBs
- Diuretics
