NSAIDs Flashcards
What enzymes do NSAIDs target?
Lipoxygenase
Cyclooxygenase
What are some pros and cons of NSAIDs?
Few side effects
Low to moderate pain
What does cox 1 do?
Platelet aggregation
Vasodilation
Gastric cytoprotectin
What does cox 2 do?
Induced by acute inflammation
Inhibited by glucocorticoids
What is prostacyclin do?
Decreased platelet aggregation
What does thromboxane do?
Platelet aggregation
What does aspirin do?
Irreversibly inhibits both cox enzymes
What are the analgesic effects of NSAIDs mediated by?
Prostaglandins e and g
How are the antipyretic effects of NSAIDs mediated?
The hypothalamus
What is the main adverse effect of NSAIDs?
Gastric bleeding by inhibiting prostaglandins and prostacyclins which help build the mucosa
What effect do NSAIDs have on clotting time?
2x increase for 7days
How do NSAIDs stimulate respiratory distress and electrolyte disturbances?
Hyperventilation
Metabolic acidosis by uncoupling oxidative phosphorylation so lactic acid accumulates
Treat with bicarbonate
What are toxic effects of aspirin?
Tinnitus
Headache
Hearing loss
How do you treat aspirin overdose?
Vomit
Sodium bicarbonate
Transfusion
Dialysis
What causes aspirin hypersensitivity?
Nasal polyps are caused
Non immune mediated
Do to shunting of AA into the leukotriene pathway
