Azithromycin, Gamithromycin
What is the 15-member ring triamilide?
Tulathromycin
What are the 4, 16- member rings?
Tylosin, Tilmicosin, Tylvalosin, Tildipirosin
Are these compounds acidic or basic? In what pH will they ionized?
Basic (7.1+)
All will ionize in acidic environment (rumen)
Are macrolides bacteristatic or bactericidal?
Normally bacteristatic but some now bactericidal
How do macrolides work?
Inhibition of protein synthesis
What is the spectrum of veterinary macrolides?
Anaerobes: +++++
Gram + aerobes: +++++
Gram - aerobes: ++
Are macrolides active against rhodococcus? Mycoplasma?
Yes
Tylosin & tulathromycin
Which macrolides are used for rhodococcus? What combination is regularly used?
Azithromycin/erythromycin-rifampin
Why is Tilmicosin not effective against mycoplasma?
Mycoplasma bovis MIC is to high & Tilmicosin is not clinically effective
What is another notable effect of macrolides unrelated to their antimicrobial activity?
Anti-inflammatory
Which macrolides have prokinetic effects on the GI?
14 (Erythromycin, clarithromycin) and 16-member rings (tylosin, tilmicosin, tylvalosin, tildipirosin)
Cross-resistance to other macrolides is common, what other antimicrobial is it common for bacteria to be resistant to if also resistant to a macrolide?
Lincosamides
What is the route of administration for Tilmicosin? Why?
SQ ONLY
IV is fatal!
IM is extremely irritating
Why should you never give systemic tilmicosin to swine?
Fatal
Is tilmicosin available as a top dress? Which animal(s)?
Yes, swine & cattle
Is oral tilmicosin the only way it can be delivered to swine?
Yes because it’s fatal
Tylosin is labeled for IM administration in what animal?
Cattle but IV and SQ use has occured
