LATG Ch 11 Gnotobiotics Flashcards
Gnotobiotics
Gnotobiotics
“well-known life”, is the study of animals whose microflora is completely defined
Germ-free, also known as axenic,
animals are only one type of gnotobiote—those free of all demonstrable microorganisms
defined flora (DF) animal.
DF animals are animals that were originally germ-free but were given a known group of bacteria and/or other organisms to aid their digestive functions
specific pathogen-free animals (SPF).
SPF animals have normal flora but are free of particular organisms, usually pathogenic, but they are not gnotobiotes.
microbiome
refers to the thousands of microbial species that inhabit a specific host.
Housing in isolators
isolators must undergo 3-6 weeks of consecutive negative testing before animals are housed in them.
Sterilization
steam autoclaving, dry heat (air), peracetic acid (2% w/v concentration), ethylene oxide, gamma irradiation, membrane filtration (sterilizing liquids by passing them through filters with pore sizes that will not permit the passage of bacteria or larger contaminants), chlorine dioxide, and hydrogen peroxide gas plasma.
What is a “Schaedler cocktail” or “altered Schaedler cocktail”
A solution containing a variety of normal flora bacteria
What is a good choice to help maintain genetic integrity when producing axenic pups by hysterectomy derivation?
Make sure that the donor female and the foster mother have a different coat color.
Which of the following represents a difference between hysterectomy derivation and embryo transfer
In hysterectomy derivation, the fetuses are at term, but in the other technique, embryos used are at an earlier stage of development.
