effect of ph on amylase Flashcards
stage 1
place 1 drop of iodine solution into each well of a spotting tile
stage 2
take 3 test tubes filled with 2cm^3in each: starch solution, amylase sol and buffer sol
stage 3 why
place all in a waterbath at 30*c
leave for 10 mins to allow the sols to reach the correct temp
stage 4
comebine all 3 sols into one test tube and mix with a stirring rod
stage 5
return mixed sol into a waterbath and start a stopwatch
stage 6
after 30s use the rod to transfer one drop of the mixed solution to an iodine well
stage 7
iodine should be blue black showing starch is present
keep taking a sample every 30s until iodine remains orange
stage 8
repeat using different pH buffers
problem: time
why
samples taken every 30s only - only approx time for reaction to complete
how can we address problem of time
take samples over shorter intervals like 10 seconds
problem: colour
why and how addressed
looking for time the iodine isnt blue black - not aways obvious
addressed by asking for others opinions
