CBC (PCV and Diff white blood cell count) Flashcards
what is a CBC
Complete blood count- groups of tests to evaluate the circulating blood
tests in a CBC
PCV, TP, WBC count, differential WBC count, RBC count, hemoglobin values, Indices (formulas)
PCV, what it stands for, what it is also known as
Packed Cell volume, Hematocrit
best test to determine RBC number or O2 carrying ability (is there enough RBCs?)
PCV/ hematocrit
tubes used for PCV test
microhematocrit tubes
difference between blue and red microhematocrit tubes
red= heparinized (has anticoagulant) take blood directly from animal blue= non-heparinized (no anticoag.) take blood from whole blood sample
how fast and how long to centrifuge d/c blood and ruminant blood
d/c= 10,000 rpm, 5 min ruminants== 10,000 rpm, 10min
the clear portion of PCV tube is what
plasma
what the thin white portion of a PCV tube is
the buffy coat (WBCs and platelets)
read hematocrit tubes at; top of rbcs or top of buffy coat
top of rbcs
normal dog PCV reading (average and range)
avg= 45% range= 37%-55%
low pcv % indicates
anemia
high pcv % indicates
dehydration
what pink plasma indicates
hemolysis
what yellow plasma is called
icterus (could be # of diseases)
what white plasma indicates
Lipemia (fat in blood) indicates that the animal ate recently
what a blood smear is used for
differential WBC count
how romanowsky staining works
based on pH of components
3 solutions in dif-quick process
fixative(green), eosin(red/orange), Methylene blue
purpose of fixative (green) solution in dif-quick procedure
preserve cells on slide, preps for stain, helps take up stain
for each solution in a dif-quick stain, how long to dip and how many times
5 times, 1 second dips
which stain is basic, which is acidic
eosin= acidic
methylene blue= basic
basic components of cells, what stain they attract?
hemoglobin, cytoplasm, granuals in some WBCs attract eosin (opposite)
acidic components of cells, what stain they attract?
nucleus (DNA), some WBC granuals
attract methylene blue (opposite)
define basophilic in terms of what color it stains
stains blue (takes up basic dye, is an acidic component)
define acidophilic/eosinophilic in terms of what color it stains
stains pink/orange (takes up acidic dye, is a basic component)
what color a neutrophilic component stains
pink+blue/grey
how new methylene blue staining works
blood mixed w methylene blue, smeared
What is new methylene blue staining used for
Quick staining and screening
