1, 2 Flashcards
Crystal violet
binds to peptidoglycan in the cell wall
Lugol’s iodine
fixes the crystal violet to the peptidoglycan
Carbol fuchsin
red stain applied so that gram negative cells can be visualised
Gram positive
blue/purple
gram negative
red/pink
first step of gram stain
crystal violet, 30 seconds, then rinse
second step of gram stain
lugol’s iodine, 30 seconds, then rinse
third step gram stain
alcohol or acetone to decolonise, then rinse
forth step gram stain
carbol fuchsin, 30 seconds then rinse
heat fixing
- Slide is passed briefly through the flame of a bunsen burner
- Cant get too hot or the bacteria will alter their shape
- Ensures specimen doesn’t wash off during staining
staph, strep and enterococcus are all
gram positive cocci that are facultative anaerobes - meaning they grow in air and anaerobically
staph morphology
round, arranges in grape like clusters
strep morphology
round arranged in chains
enterococcus morphology
short chains or pairs
the catalase test differentiates between
staph vs (strep/entero)
