What is MOA_heparin and LMWH?
Interfere with the clotting factor activation in both the intrinsic and extrinsic pathways.
Bind to antithrombin III with the subsequent rapid inactivation of coagulation factors esp thrombin and Xa.
What are classes of Anticoagulant drugs?
- Heparin and LMWH (Low molecular weight heparin)
- Oral anticoagulants
- Thrombin inhibitors
What is LMWHs?
Extracted from heparin that have larger anti-factor activities, greater bioavailability, longer half-life than heparin.
What is the major adverse effect of anticoagulant drugs?
Hemorrhage
What is the antidote for heparin poisoning?
Protamine
What is MOA_oral anticoagulants such as warfarin (coumadin)?
Antagonize vit. K, interfere with the synthesis of vit K-dependent clotting factors (II, VII, IX, X)
What is the antidote for oral anticoagulant?
administration of vit K can overcome the anticoagulant effects of the oral anticoagulants, but the effect takes about 24 hours.
