A
- These drugs block ATP-sensitive K+ channels, resulting in depolarization and calcium influx in β-cells. Increased [Ca]i stimulates insulin release.
- Also decreases hepatic glucose production and improves insulin sensitivity.

- Sulfonylureas: Duration of action ranges from 12 to 24 hours.
- Glinides: Faster onset and shorter duration of action than the sulfonylureas
