SHOULD BE IN HEMA XXXRETICULOCYTE COUNT Flashcards
To detect the presence of RNA, the red blood cells must be stained while they are living and this process is called
supravital staining
GIVE THE ORDER OF ERYTHOCYTE FORMATION STARTING FROM PRONORMOBLAST
PRONORMOBLAST
BASOPHILIC NORMOBLAST
POLYCHROMATOPHILIC NOMOBLAST
ORTHOCHROMATIC NORMOBLAST
POLYCHROMATOPHILIC ERYTHROCYTE
ERYTHROCYTE
The number of reticulocytes in how many red blood cells is determined.
1,000
what are the materials used in reticulocyte count
glass slides
anticoagulated blood - edta
supravital stain
automatic pipette
microscope
cedarwood oil
procedure for reticulocyte count
Place three drops of reticulocytes stain in a small test tube.
Add three drops of well mixed whole blood to the tube containing the stain.
Mix the tube and allow to stand at room temperature or incubate at 37C for 15 minutes
After 15 minutes, mix the contents of the tube well and prepare smears and allow it to dry.
Examine the smear under the oil immersion objective.
Count the total number of reticulocytes found per 1,000 RBCs
formula fpr reticulocyte count
RETICULOCYTE COUNT (%) = (Number of Reticulocytes Counted / 1,000) x 100
reference range for retics count
Adult: 0.5-1.5%
Newborn: 2.0-6.0%
why do we need to use supravital stains?
dyeing while the cells are in the living state
what is the significance of the reticulocyte count
it assesses the erythropoietic activity of the bone marrow to detect underlying diseases such as thalassemia or aplastic anemia
what are the 3stains used in supravital staining of retics?
New methylene blue
Pure azure blue
Brilliant cresyl blue
the most common among the supravital stain as it stains the retics deeply and uniform
NEW METHYLENE BLUE
substitute or 2nd option if the new methylene blue is not avaiablable
pure azure blue
least used supravital stain for retics
Brilliant cresyl blue
upon staining using supravital the rbc will appear what color?
bluish green
upon staining using supravital the rna of the retics will appear
blue
why edta is the ideal tube top?
it is stable for the span of 24 hrs, however, the retics will decrease within 6-8 hrs after collection
3 components that are sometimes being misidentified as retics
pappenheimer
heinz bodies
howell jolly
why papperheimer is being misidentified as a retic?
because of the presence of iron that appears like rna after staining
why heinz bodies are being misidentified as a retic/
it composed of denatured rrbc
why howell jolly is being identified as a retic
it has the remnants of rna
what are the 2 interferences of retics count?
heparin and the high blood glucose level
why high blood glucose level is included in the interferences of retics count?
will cause pale staining to the supravital stain
conditions where retics are found elevated or high.
- hemolytic anemia
- IDA
- thalassemia
- sideroblastic anemia
conditions where retics are found low or decrease
- aplastic anemia
- BM failure
- kidney failure
