B lactam ring looks like D-Ala-D-Ala, which means that the enzyme of the bacteria will bind to the beta-lactam ring, and the enzyme is never functional again after this. Because of this peptidoglycan (a mesh network which gives rigidity to bacteria) is not formed and the bacteria grow to their death.
