Why does Alzheimer’s disease start with memory loss?
Because the shrinkage of the brain starts in the hippocampus, where the memories are stored.
What do plaques contain in Alzheimer’s disease?
A small peptide called amyloid beta, this is aggregating and is very toxic when it aggregates.
What is APP? (explain)
A mutation that can lead to the generation of amyloid plaques.
What does the tau protein normally do?
Normally tau binds microtubules and stabilises them, tau can also phosphorylate to let go of the microtubules.
How is tau affected in Alzheimer’s disease?
The tau protein gets hyperphosphorylated and aggregates, and then becomes tangles.
What is the amyloid cascade hypothesis?
Mutations in APP or APP processing enzymes lead to tau pathology, which leads to neurodegeneration.
