Lipopeptides Flashcards
What is the lipopeptide we talked about?
Daptomycin
Daptomycin Mechanism of Action
Binds bacterial membranes and causes depolarization of membrane potential
Inhibits DNA, RNA, and protein synthesis
Daptomycin exhibits ___ dependent ____ activity.
Concentration dependent bactericidal activity
Daptomycin Spectrum of Activity
Gram Positives only
MSSA, MRSA, VRSA
PRSP
VRE
Daptomycin Absorption and Distribution
Only given by IV
Rapidly distributes to well perfused tissues
Daptomycin Elimination
Primarily excreted renally.
Adjust dose for renal insufficiency
Daptomycin Clinical Uses (and when not to use)
Used for serious/complicated infections by resistant bacteria
Should NOT be used in pneumonia
Daptomycin Adverse Effects
Myopathy and CPK Elevation in small %
GI - nausea/diarrhea
HA
Rash
Daptomycin Drug Interactions
Interact with HMG-CoA reductase inhibitors to cause increased risk of myopathy
Quinipristin-Dalfopristin
Have a synergistic effect as protein synthesis inhibitors for Gram Positive organisms
