DNA synthesis Inhibitors Flashcards
Class I DNA Synthesis Inhibitors
Antifolaxe drugs
-sulonamide and trimethoprim
Sulfonamide
- mechansim
- names + uses
- Side FX
PABA analog that inhibits Dihydropteroate synthase
Bacteriostatic @ singly, bactericidal @ DHFR inhibitor
Sulfa/sulfi drugs-usually UTI but sulfasalazine is for IBD
precipitate in urine, cross allergenic, fever, rash, NVD
Tripemhoprim
- mech
- use
Inhibits DHFR (DHF to THF) with specify to bacterial
Acute UTI
Bacterium
wide variety use-shigella, salmonella, UTI, prostates, non-TB mycobacteria
MSSA, MRSA, respiratory tract infections (Haemophilis, Klebisiella)
Gram - sepsis including MDR (enterobacter and serratia)
Class 2
- mech
- how know drug is in this class
- use
-DNA Gyrase (prevents release of supercoiled DNA)/Topo IV (can’t separate replicated DNA @ cell division)inhibitor-block bacterial DNA synthesis…allow cut but not reseal
flox or just ox
bactericidal vs gram- (gyrase) and gm + (topo IV)
Class 2 DNA Synthesis Inhibitors
-major division + names (general)
-clinical uses (of major one and which are resp)
Side effects
-Great Gm- + moderate Gm+- (Cipro and all others except MG…see below)
vs Improved Gm+ (broad spectrum) Gemifoloxacin and mooxifloxacin
- Cipro best for Gm-, Anthrax, MDR UTI, Diarrhea (Shigella, Salmonella, ETEC, Camplylocater), soft tissue/bone/joint/respiratry infections
- -respitory=levofloxacin, gemifloxacin and moxifoxacin
-Rare, NVD, reversible arthopathy, DAMAGE GROWING CARTILAGE
