Cell Structure and Microscopes Flashcards
What level of detail of a cell can you see using a light microscope?
Minimal detail, the rough outline of a cell and maybe the nucleus and the outline of unidentifiable organelles
Nile blue stain can stain which cell feature?
The nucleus
Are ribosomes found in prokaryotic cells or eukaryotic cells?
both
Does a prokaryotic cell contain RER?
No
Does a prokaryotic cell contain a golgi apparatus?
No
What are prokaryotic cell walls made out of?
Peptidoglycan
Do prokaryotic cells have extrachromosomal DNA that is circular?
Yes
What size ribosomes do prokaryotic cells have?
70S
Is RER and/or SER found in both plants and animals cells?
both are found in both
Is a ribosome a membrane-bound organelle?
No
Name of feature which is common to bacteria, fungi and protoctista
ribosomes
What is the maximum resolution of a laser scanning confocal microscope?
200 nm
Is a laser scanning confocal microscope 2D or 3D?
either
What is the image colour of a laser scanning confocal microscope?
coloured
What is the maximum resolution of a scanning electron microscope?
3 - 10 nm
Does a scanning electron microscope produce 3D or 2D images?
3D
What is the image colour of a scanning electron microscope?
black and white
What is the maximum resolution of a transmission electron microscope?
0.5 nm
Does a transmission electron microscope produce 2D or 3D images?
2D
What is the image colour of a transmission electron microscope?
black and white
Outline how a student could prepare a temporary mount of tissue for a light microscope
1, Obtain thin section of tissue e.g. using ultratome or by maceration
2, Place plant tissue in a drop of water
3, Stain tissue on a slide to make structures visible
4, Add coverslip using mounted needle at 45 degrees to avoid trapping air bubbles
Functions of the golgi apparatus
transport, sorting and modification of both proteins and lipids
Cells in culture will absorb amino acids from the surrounding culture medium. If radioactively labelled glycine is provided in the culture medium, in which organelle will radioactivity be found first?
ribosomes
Intestinal epithelial cells have observable “brush borders” seen on the cells. What are they?
microvilli
When mitochondria are extracted from cells, they are usually kept in a 0.25 moldm-3 sucrose solution. What is the purpose of the sucrose solution?
it prevents the mitochondria from changing structure
Which one of the following has the lowest surface area to volume ratio? (virus, phagocyte, red blood cell, bacterium)
a phagocyte
Are centrioles found in plants?
No
