GP IIb Receptor antagonists include
Abciximab, Eptifibatide, Tirofiban
Abciximab - MoA
binds to BOTH activated and non-activated platelets
Eptifibatide & Tirofiban - MoA
mediate binding of fibrinogen to the receptor complex. Activated platelets only
GP IIb antagonist toxicities
bleeding and thrombocytopenia
What is the IIb/IIIa complex?
a platelet membrane receptor for fibrinogen and vibronectin/other clotting factors
PAR 1 antagonists include
vorapaxar (zontivity)
Par-1 antagonist indications
approved for oral use to reduce risk of thrombotic cardiovascular events in patients with history of MI or artery disease
Vorapaxar MoA
blocks PAR-1, the major thrombin receptor on platelets - inhibits aggregation
Vorapaxar toxicity
bleeding, anemia, depression, rash
Vorapaxar contraindications
history of stroke, intracranial , intracranial hemorrhage shouldn’t be used with CYP3A4 inhibitors OR inducers
Dipyridamole indications
used in combo to prevent cerebrovascular ischemia, also used with warfarin for prevention of embolus
