6: Pathways of Cell Signaling Flashcards
Growth factor receptor kinases
extracellular growth factor binds to receptor causing a conformation change and stimulation of kinase activity, the phosphate groups on the kinase attracts other proteins
What is an example of a growth factor receptor kinase?
insulin
Ras-activating protein
helps exchange GDP for GTP, making Ras active
Ras
monomer becomes active when bound to GTP, activating more signaling pathways
MAP
mitogen activated protein
What enzymes does Ras activate?
- MAP-kinase-kinase-kinase
- MAP kinase-kinase
- MAP kinase
What does MAP-kinase signal?
prepare for mitosis
Epidermal growth factor (EGF) receptor
example of tyrosine-kinase receptor; EGF binds to an extracellular ligand domain, activating many pathways
What do steroidal anti-inflammatory drugs block?
intracellular recpetor
What hormones work via an intracellular receptor?
cortisol, sex hormone, thyroid hormone
TGF-beta
transforming growth factor-beta
SMADs
recruited to a receptor that has been activated by TGF-beta and heads to the nucleus to activate a gene promoter
CREB
cAMP response element binding protein
What does CREB do?
it is a target of protein kinase that activated gene expression
What phosphorylates CREB?
active protein-kinase A
