Q

What are the alternative for Lauryl tryptose broth?

A

Lauryl sulfate broth

If none

Lactose broth

17
Q

What is the tube that is inverted in Lauryl tryptose broth?

A

Durham tube

18
Q

How to determine the LTB strenght?

A

By adding water (100, 20, 10 mL) to the medium

19
Q

What is the ratio for Triple strength LTB for sample?

A

10mL Triple strength LTB for 20mL of sample

20
Q

Ex. 5g/10mL + 20 mL sample = 5g/30mL (Need adjustment )

15g/10mL + 20mL sample = 15g/30mL

A
21
Q

Procedure for Presumptive in MTFT?

A
  1. Use Lauryl tryptose broth then determine the Triple strength of LTB
  2. Arrange fermentation tubes in row of five
  3. Mix the sample by shaking the bottle for about 25 times
  4. Inoculate each tube with 20 mL water sample portions
22

  5. Mix test portions in the medium by gentle agitation
  6. Incubate at 35 +- 0.5 (24 hrs: turbid, gas and acid reaction / 48 hrs)
  7. Examine the turbidity, gas production and acidic reaction
23
Q

What is the positive indicator for Multiple tube fermentation technique?

A

Presence of growth, Acid, and gas

24
Q

NOTE: Tubes that are Positive are submitted to the confirmed phase

A
25
Q

What media is used in confirmed test?

A

Brilliant green lactose bile broth with inverted durham tube

26
Q

What is the pH for Brilliant green lactose bile broth?

A

pH 7.2+-0.2

27
Q

Process for confirmed phase?

A
  1. Use BGLB with inverted durham tube
  2. Gently shake the tubes to re-suspend the organism
  3. Transfer one loopful of the culture from LTB to BGLB broth
  4. Do this in all presumptive test
  5. incubate at 35+-0.5C
  6. Check the result and calculate the MPN value those who are positive
28

What is the size in diameter for sterile loop?

A

3.0 - 3.5 mm

29
Q

Positive indicator for Confirmed phase?

A

Formation of gas or turbidity within 48 hrs +- 3 hrs

30

A

EC (E. coli) broth / EC-MUG

31
Q

What is the ph for EC broth?

A

6.9+-0.2

32
Q

NOTE: The process of Completed phase is similar to Confirmed phase (Except incubation)

A
33
