Pharmacology and presentation of Valproate?
400mg powder ampoule
-Anticonvulsant, that predominantly blocks sodium channels but also enhances the activity of GABA at it’s receptors within the central nervous system.
Indicators of Valproate?
- Status epilepticus that has not responded to two doses of midazolam
Contraindications of Valproate?
- Known severe allergy.
Cautions of Valproate?
- None
Side Effects of Valproate?
- None
Dosing Regimes of Valproate?
Administer IV, dissolve with 4ml of saline, dilute to 10-20ml
- If giving Valproate ICP required
Adult IV
-1200mg IV, once
Paediatric IV or 300mg per 10kg IV once except for 30kg
- 5kg = 150mg IV, once
- 10kg = 300mg IV, once
- 20kg = 600mg IV, once
- 30kg = 800mg IV, once
- 40kg = 1200mg IV, once
Onset and duration of Valproate?
Onset:
- 10-20mins
Duration
- 6-12hrs
