Describe process
A
- agar gel set in a Petri dish
= bacteria grow to visible colonies

How to avoid risks
A
- sterilise all Petri dishes
= kills unwanted microorganisms and prevents contamination - bacteria transferring by using inoculating loop
= sterilise it before use through a Bunsen burner flame - once bacteria transferred
= use a adhesive tape
= stop lid from falling off and unwanted microorganisms entering - place agar plate upside down
= stops moisture from dripping down into bacteria and disrupting colonies

Temperature of incubation
A
Normally incubate at 25C
= reduce changes that harmful bacteria will grow

Process of effects of antibiotics on bacterial growth
A
- clean bench with disinfectant
- sterilise inoculating loop through flame
- open sterile agar gel plate near flame
= flame kills bacteria in the air - use loop to spread chosen bacteria evenly over plate
- place sterile filter paper discs containing antibiotic onto plate
- incubate plate at 25C
