sildenafil, vardenafil
A
- PDE5 inhibitors, increase cGMP, smooth muscle relaxation in the corpus cavernosum, inc blood flow, penile erection
- tox: headache, flushing, dyspepsia, impaired blue-green color vision. do not take with nitrates

leuprolide
A
GnRH analog with agonist/antagonist properties
- pulsatile used for infertility
- continuous used for prostate cancer, precocious puberty, uterine fibroids
- tox: antiandrogen, n/v

- bind estrogen receptors
- used for hypogonadism or ovarian failure, menstrual abnormalities, HRT, men with androgen-dependent prostate cancer
- tox: inc risk of endometrial cancer, bleeding in post menopausal women, clear cell carcinoma with DES in utero, inc risk of thrombi
- DO NOT GIVE with ER + breast cancers, or history of DVTs

- antagonist at estrogen receptors in hypothalamus (prevents normal feedback inhibition) and inc release of LH and FSH from pit
- used for infertility due to anovulation (PCOS)
- may cause hot flashes, ovarian enlargement, multiple simultaneous pregnancies, visual disturbances

- SERM: antagonist on breast tissue, agonist on bone, uterus
- associated with endometrial cancer and DVTs
- used to treat/prevent recurrence of ER+ breast cancer

- increased risk of VTE, decreased resorption of bone
- used to treat osteoporosis
