Morphine
A
Strong agonist to mu receptors. Decrease cAMP = decrease pain

A
100 x more potent that morphine. Can be used a sedative

S/E morphine
A
Decrease sensitivity to CO2 in respiratory centre = respiratory depression

Codeine
A
Moderate agonist. Broken down into morphine by CYP2D6

Issues with codeine
A
SSRI (fluox) inhibits breakdown. Variable expression dependent of race etc. Constipation.

Buprenophine
A
Ant/agonist. Very high affinity for mu. Partial agonist

S/E buprenophine
A
Low BP, nausea, dizziness, treat addiction, not easily reversed.

Naloxone
A
Reversal agent. High affinity for mu, so displaces morphine.
