MOA of medications Flashcards
How does Memantine work?
NMDA antagonist slowing influx of calcium which halts cell destruction
How do benzos work?
They enhance the effect of GABA at teh GABA receptor
MAOis work on both MAO-A and MAO-B, which works selectively on MAO-B at lower doses?
Selegiline
What does MAO-A metabolize?
serotonin and norepinephrine, dopamine
tyramine in the gut
What does MAO-B metabolize?
phenylethylamine, dopamine
how does sildenafil, vardenafil, and tadalafil work?
nonselective phosphodiesterase inhibitors
- cannot be used with NO agonist (nitrates)
ramelteon
melatonin agonist in suprachiasmatic nucleus of hypothalamus
8mg qhs
z-drugs work at what recptor
benzo receptor agonist
zaleplon (sonata)
shortest half life 1 hour
compared to ramelteon (2.5/5), zolpidem (2.5-4.5), eszopiclone (6 hrs)
-useful for middle of the night insomnia
naloxone
opioid antagonist
naltrexone
opioid antagnoist w/ half life of 72 hours
buprenorphine
mixed opioid agonist-antagonist
bromocriptine
mixed dopamine agonist-antagonist
used for parkinsons disease
riluzole
glutamate antagonist
FDA approved for ALS
-GI upset, fatigue, dizniness, liver enzyme elevation
tiagabine
gaba reuptake inhibitor
- adjunctive treatment for partial complex seizures
memantine
NMDA antagonist FDA approved for treatment of moderate to sever Alzheimers dementia
GABA-A receptor is waht type of ion channel
chloride
how does mirtazapine work?
blocking presynaptic alpha 2 receptors which stops feedback inhibition on the release of NE into the synaptic cleft. results in more NE released
- antagonism of presynaptic alpha 2 receptors leads to potentiation of serotonergic and noradrenergic neurotransmission
- lacks anticholergic side effects, lacks sexual side effects, lacks anxiogenic side effects
MAO of triptans
agonist at 5HT 1B and 5HT1D
- modulate the excitability of cells in the trigeminal nucleus caudalis
Mirtazapine has 5HT3 antagonism what is this thought to be responsible for?
antinausea
