Macrolides prototype
Erythromycin (Macrolides are used mainly for endocarditis prophylaxis before dental procedures).
Erythromycin indications
Treatment of respiratory, dermatological, urinary tract, and gastrointestinal infections caused by susceptible strains of bacteria.
Erythromycin actions
Binds to cell membranes, causing a change in protein function and cell death; can be bacteriostatic or bactericidal.
Erythromycin route
Oral, 1-2 h, 1-4 h;
IV, rapid, 1 h
Erythromycin T 1/2
3 to 5 hours; metabolized int he liver, excreted in bile and urine.
Erythromycin adverse effects
Abdominal cramping, vomiting, diarrhea, rash, superinfection, liver toxicity, risk for pseudomembranous colitis, potential for hearing loss.
