lab quiz Flashcards
competent cells
can pick up plasmids easier
x-gal
special (blue) food that turns the cell blue
IPTG
up regulate lac Z gene (makes more, so blue color is more obvious)
heat shock
open cell walls by briefly heating
transformation
inserting plasmid back into bacterial cell/host
blue white color screening
allows us to differentiate cell with vs. without plasmid
lac Z allows cells to turn
blue (beta-galactosidase)
if lac Z does not work,
it means our gene of interest was probably inserted at the right location
why do we want the white colonies?
it means Hind and Eco cut at the right spot (it cut out the lacZ) and inserted our DNA into that location
what happens to bacteria without plasmid in AMP (antibiotic)?
the bacteria without the plasmid will not have AMP, so it will die.
what did we use as our heat shock cells?
E coli cells
