Lab 1 Flashcards
Aims of bacterial culture
- Isolation of causative agents
- AB susceptibility test
- Vacc. prod.
- Industry
Types of detection of bacteria in a sample
- Direct: smear, impression smear or PCR
2. Indirect: animal trial or inoculation of sample into a medium
Composition of media
- Water
- C-sources
- N-sources
- Vitamins and other additives
- Osmotic pressure: 0,9% NaCl sol.
- pH: 7.2-7.4
Classification of media
Accordring to:
- Origin: natural/artificial/synthetic
- State: liquid/semi-solid/solid
- Aim of culture: common (basic nutrient)/selective/differential (indicator)
Natural media examples
Potato slide, blood, serum, milk, bile, urine
Common media is
Capable of sustaining growth of bacteria
Selective media is
Contain inhibitory substances that prevent growth of unwanted bacteria sp.
Differential media is
- Give a presumptive ID of bacteria
- Often contain sugar + pH-indicator (colour change)
Semi-solid medium examples
- Motility tests
- Oxidation/fermentation
Solid medium examples
- Blood agar
- Nutrient agar
- Mac Conkey´s agar
Liquid medium examples
-Carbohydrate broths
Blood agar is
Solid, artificial, differential medium
Nutrient agar is
Solid, artificial, common medium
Carbohydrate broths is
- Mannitol/Dextrose/Lactose/Sucrose
- Liquid, artificial, differential medium
- pH-indicator: phenol-red
Mac Conkey´s agar is
- Solid, artificial, selective, differential medium
- Lactose + (e-coli) or - (salmonella)
- Inhib. materials: bile salts, crystal violet
- pH-indic: neutral red
