LAB 1 Flashcards
What does dilution factor 10 mean `
1 part solution
9 parts buffer
10 fold, 10 times, 1in10 dilution
Where does iodine bind to startch
Amylose
Why does pH damage protein structure
Leads to the protonation of ionizable side chains leading to more exsposed hydrophobic cores and aggregation together through hydrophobic interaction
What is beer’s law
Concentration of a solution is linearly proportinonal to the absorbtion of light
What is lamberts law
Lamberts law is that the intensity of transmitted light is proportonal to the pathlength
How is an absorbance spectra made
Made by increasing wavelength and plotting absorbtion as it increases
What is the beer lambert equasion and units of components
Absorbtion (no unit) = ε (L/mol*cm) * c (mol/L) * pathlegth (cm)
What is the name of the formation of beta sheet + alpha helix
Nucleation
What is the specific porphyrin rings found in heme
Fe-protoporphyrin IX.
What are the axis of an absorbance spectra, what does this measure
x axis = Absorbtion
y axis = wavelength
Used to determine the maximum absorbance of a molecule -> will be the highest peak of the absorbance spectra
What are the axis of a progress curve and what is this used for
x axis = product/reactant - which can be measured by their absorption
y axis = time
Used to determine the Vi of a reaction as a given enzyme conc and substrate concentration
What are the axis of a michales mention graph and it used for
X axis = [s]
y axis = [v]
Used to determine properties of the enzymes such as km and vmax
What are the axis of an enzyme assay and it used for
x axis = concentration
y axis = inital rate of reaction
NOTE: this is the same as a michalis mention graph
