Ketoconazole Flashcards
Class
Fungal lanosine
demethylase inhibitor
Actions
Antifungal
Mechanism of action
increases fungal cellular permeability
Clinical use
Antifungal agent used for long periods at high doses especially in immunosuppressed patients
Adverse effects
Liver injuries and
adrenal gland problems
What does Ketoconazole inhibit
14-α demethylase
what is Ketoconazole
: A broad spectrum anti-fungal agent (A Lanosine demethylase inhibitor)
what does 14-α demethylase do
, a cytochrome P-450 enzyme which converts lanosterol to ergosterol
what is a result of the ergosterol inhibition
causes increased fungal cellular permeability
other possible mechanisms for ketoconazole action
1) Inhibition of endogenous respiration.
2) Interaction with membrane phospholipids.
3) Inhibition of yeast transformation to mycelial forms.
4) Inhibition of purine uptake.
5) Impairment of triglyceride and/or phospholipid biosynthesis.
