HIV Flashcards
HIV classification and characteristics:
o Genome: 2 copies of positive polarity RNA
o Capsid symmetry: Icosahedral
o Envelope: Yes; includes gp120 and gp41 (mediate interaction between viral partical and host cell)
o Tropism: CD4+ T cells (also CD4+ monocytes, macrophages)
o Reverse transcriptase + integrase + protease = 3 E that come with the virus
capsid protein detectable by ELISA for dx:
p24
3 viral enzymes:
Reverse transcriptase
integrase
protease
2 envelope glycoproteins:
gp 120
gp 41
viral protein that binds CD4:
gp 120
viral protein binds chemokine R, initiates fusion with host plasma membrane
gp 41
What does reverse transcriptase do?
Where does it do it?
ssRNA –> ssDNA –> dsDNA
cytoplasm
Enzyme that gets viral dsDNA into host DNA?
integrase
last viral enzyme used to package new viruses?
protease
what makes the new viral RNA and mRNA?
host RNA pol 2
How is HIV tested for?
ELISA for anti-p24 antibodies
then
confirm with Western Blot
which two enzymes are required for formation of the provirus?
reverse transcriptase
integrase
major cause of CD4 cell loss in HIV?
binding of viral envelope proteins expressed on infected cell surface binds to CXCR4 –> autophagy
co-receptors for viral binding to host cell:
CXCR4
CCR5
gag?
capsid proteins
ie. p24
