disulphide bridges - fairly dtrong and therefore not easily broken

place sdample solution in a test tube and add an equal vol of sodium hydroxide solution at room temp

add after drops of of very dilute copper sulfate solution and mix gently

a purple solution indicates the presence of peptide bond, if no protein is detected then the solution remains blue
