Enzyme: how fast product is made - Required Practicals Flashcards
1
Q
What does catalase break down
A
H2O2 → H2O + O2
2
Q
As oxygen is produced in the reaction where does the water go
A
The oxygen displaces the water in the measuring cylinder
3
Q
Outline method for measuring rate of enzyme action for catalase
A
- Set up boiling tubes with the same vol and conc of H2O2 and another set of boiling tubes with catalase in.
- Add equal volumes of buffer to each test tube
- Set up apparatus: trough of water, upturned measuring cylinder, delivery tube held by clamp.
- Put each boiling tube and another set of boiling tubes with catalase set to 10 - 40
- Wait 5 mins
- Pipette same volume of catalase to each of the boiling tubes
- Attach bung and delivery tube to test tube
- Record more oxygen is produced in first minute using stop watch
- Repeat and calculate average amount produced
- Calculate rate
4
Q
What would a negative control be in this experiment
A
A boiling tube containing without adding catalase for each temp
5
Q
Why is a buffer used
A
Keep ph constant
6
Q
Why is the catalase also put into the water bath
A
To see if the enzyme is denatured by temp
