Beta oxidation of fatty acids Flashcards
When dogs were fed odd-chain length fatty acids with phenyl groups at the methyl end, what was consistent about the molecule that was excreted?
It was always benzoate
When dogs were fed even-chain length fatty acids with phenyl groups at the methyl end, what was consistent about the molecule that was excreted?
It was always phenyl acetate (benzoate w/ a CH2 between the benzene and the COO- group)
What is a beta carbon in Knoop’s experiment?
One that is the second carbon away from the COO-
What are the three stages of beta oxidation of fatty acids?
Activation, transport and oxidation
What is a through lne?
Modification of starting material such that subsequent reactions have a negative deltaG
First step of Activation?
The fatty acid attaches to coenzyme-A
What is formed as a result of a FA attaching to coenzyme A?
Acyl-CoA
Where does the formation of Acyl-CoA occur?
Outer surface of the mitochondria
What is the formation of Acyl-CoA catalysed by?
Acyl-CoA synthase
What are the reactants of the formation of Acyl-CoA?
RCOO- (FA), CoA, ATP
What are the products of the formation of Acyl-CoA?
Acyl-CoA, AMP, PPI
During the formation of Acyl-CoA, which product is removed?
Pyrophosphatase (PPi)
Why is PPi removed from the reactants during the formation of Acyl-CoA?
To prevent the reverse reaction from occuring
Why would you want the reverse reaction of the formation of Acyl-CoA to not happen?
If it did happen, some Acyl-CoA would be lost
How is PPi removed from the formation of Acyl-CoA?
Water is used to hydrolyse the PPi into 2 Pi and 2H+
