What is senescence?
Age dependent decline in vital physiological functions
What is the disposable soma theory?
Natural selection has caused organisms to have sufficient resources/ repair mechanisms to prevent ageing- at least until the organisms have reproduced and cared for their young
What are the senescence factors?
- Metabolism
- Reactive oxygen species
- DNA damage
What are factors that increase lifespan?
- Dietary restriction
- Environmental stress
- Signals from the somatic gonad
What is the reactive oxygen species?
O2-
What causes progeria syndromes?
Nuclear lamina mutations
What causes nestor guillermo?
BANF1 mutations- affects the nuclear lamina/ mitosis
What does diminished insulin signalling in c. elegans and drosophila cause?
Increase in lifespan
How does IGF signalling influence DAF16/ FOXO?
Inhibits FOXO
What does decreased TOR activity cause?
Increased lifespan
What are the genes downstream of FOXO?
- Antioxidant genes
- Chaperones
- Metabolic genes
- Antibacterial genes
What do sirtuins do?
Prevent ageing
They activate FOXO and the mitochondrial unfolded protein response
What activated sirtuins?
NAD- levels decline with age
