3-9 Flashcards
what is a glycocalyx?
external structure around the cell wall produced by the bacterium
what is a slime layer?
term used for glycocalyx that is loosely associated with the cell wall of bacterium
what is a capsule?
term used for a glycocalyx tightly associated with cell wall of bacterium
what is the glycocalyx (slime layer and capsule) composed of?
mucoid polysaccharides or polypeptides that repel most stains
in capsule stain what does Congo red do?
stain the slide
in capsule stain what does Maneval’s stain stain?
cell
what stains the capsule?
capsule remains unstained
appears as while halo around cell
what are the functions of the capsule?
- avoidance of phagocytosis
- reduces desiccation
- protects against certain chemicals
- aids in adherence
- serves as nutrient source during starvation
what is klebsiella pneumonia?
gram -
primary pneumonia is characterized by sudden onset and gelatinous reddish brown sputum (phlegm) (MO grows rapidly destroying lung tissue causing death)
secondary pneumonia is a nosocomial (hospital acquired) disease
what is the most common infection caused by klebsiella
pneumonia
steps of capsule staining?
- add drop of Congo red
- add MO and mix
- take second slide and slide over slide with Congo Red
- air dry (DO NOT HEAT FIX)
- flood slide with Maneval’s stain
- gently blot dry
why do we not heat fix?
destroy capsule look like false negative
shrink cell look like false positive
acidic stain
basic stain
acidic stain: stains slide
basic stain: stain cell
what are the results of capsule staining?
klebsiella pneumonia + contains capsule
Staphylococcus aureus - no capsule
