1-3 Flashcards
What is aseptic technique?
the process of handling sterile materials in a manner that prevents their contamination.
This technique also involves the use of bacteria cultures without contaminating them as well.
What is contamination?
a situation where unwanted MO(s) end up in sterile materials or cultures due to lack of proper aseptic technique
how do you label materials?
initials
lab section
table #
MO
how and where do we dispose test tubes/dishes?
remove labels
disposal cart–located next to lab door, front left of room
when not in use, where do we place media tubes?
test tube rack
do not lay tubes on table
how does one hold the inoculating needle/loop?
like a pencil (at an angle)
in dominant hand
why are petri dishes incubated upside down in the incubator?
- Prevent condensation from forming
2. Prevent contamination
How do you remove the cap off a test tube?
tucked under pinky
never placed on table
never hold tube by its cap b/c cap is loose
after removing the cap, what dos one do to the tube?
Flame the tube
how does one use the lid of a petri dish?
as a shield when inoculating on a table
what is a culture?
a medium that contains living microbes
what is a pure culture?
when a culture contains a single species
how does one adjust the Bunsen burner?
flame has an inner and outer cone
sterilization of inoculating instruments is done in the hottest part of flame (tip of inner corner)
where does one label a petri dish?
on their base, not lid
what are broths used for?
grow microbes when fresh cultures or large numbers of cells are required
